以大豆绥农28为材料,用发根农杆菌(Agrobacterium rhizogenes)K599浸染转化后诱导产生发根,通过β-葡聚糖苷酶(GUS)染色进行效率检测,确定毛状根转化的最佳条件。结果表明绥农28大豆品种发根诱导高效转化的条件如下:在1/2MSB5+20g/L蔗糖+琼脂7g/L,p H5.8萌发培养基上萌发5~6d的大豆子叶节为外植体,当菌液侵染时间为20min,共培养基p H5.4的条件时,发根转化率为58.57%。
Soybean Suinong28 was used as raw material and Agrobacterium rhizogenes K599 was used for transformation to induce hairy root formation.
