目的观察EZH2在不同卵巢组织中的表达,探讨其在肿瘤发展中可能发挥的作用。方法采用免疫组织化学染色检测15例正常卵巢组织、32例卵巢良性肿瘤、28例卵巢交界性肿瘤及50例卵巢癌中EZH2蛋白的表达,其中15例正常卵巢组织、23例卵巢良性肿瘤、23例交界性肿瘤及25例卵巢癌行RT-PCR检测。结果 1EZH2蛋白在卵巢癌细胞核中的表达明显高于正常卵巢组织(P<0.01)、卵巢良性肿瘤(P<0.01)及交界性肿瘤(P<0.05),且与肿瘤的分级和分期具有相关性(P<0.01,P<0.05);2 EZH2蛋白在卵巢肿瘤组织的细胞质中亦有表达,且良性肿瘤和交界性肿瘤中的表达高于卵巢癌(P均<0.05);3 EZH2蛋白在卵巢癌细胞核中的表达与Ki-67呈正相关性(r=0.40,P<0.01)。结论卵巢肿瘤组织细胞核及细胞质中均有EZH2蛋白的表达,且卵巢癌细胞核中EZH2蛋白的表达与肿瘤的分期及分级具有相关性,并参与细胞增殖。
Objective To observe the expression of EZH2 in different ovarian tissues and to explore its possible role in tumor development. Methods Immunohistochemical staining was used to detect the expression of EZH2 protein in 15 cases of normal ovarian tissue, 32 cases of ovarian benign tumor, 28 cases of ovarian borderline tumor and 50 cases of ovarian cancer. Among them, 15 cases of normal ovarian tissue, 23 cases of ovarian benign tumor, 23 Cases of borderline tumors and 25 cases of ovarian cancer were detected by RT-PCR. Results The expression of 1EZH2 protein in ovarian cancer cell nucleus was significantly higher than that in normal ovarian tissue (P <0.01), benign ovarian tumor (P <0.01) and borderline tumor (P <0.05), and was correlated with tumor grade and stage (P <0.01, P <0.05) .2 The expression of EZH2 protein was also found in the cytoplasm of ovarian tumors, and the expression of EZH2 protein was higher in ovarian cancer than in benign tumors and borderline tumors (all P <0.05) The expression of Ki-67 in cancer cell nucleus was positively correlated (r = 0.40, P <0.01). Conclusions The expression of EZH2 protein in both nucleus and cytoplasm of ovarian cancer tissues is correlated with the expression of EZH2 protein in ovarian cancer cell nuclei and the staging and grading of tumor.
