目的用基因芯片研究不同程度苯中毒细胞凋亡相关基因的差异表达。方法选取7名诊断为不同程度接苯中毒的工人(疑似1例、轻度2例、中度2例和重度2例)并设年龄、工龄匹配的健康者做对照,从其外周血中分离单个核细胞,用Trizol试剂盒抽提总RNA,纯化、逆转录生成cDNA产物,分别用荧光染料Cy3和Cy5标记后与含177条细胞凋亡相关基因的微阵列芯片进行杂交。结果共有41条细胞凋亡相关的基因表达异常,其中有3条基因的表达在轻、中度苯中度工人中一致性上调,1条基因在所有苯中毒工人中均表达上调,随苯中毒程度加重,细胞凋亡相关基因差异表达总数有减少趋势。结论苯中毒病人细胞凋亡相关基因的差异表达参与了苯血液毒性的发生、发展。
Objective To study the differential expression of apoptosis-related genes in benzene poisoning cells with different degrees of gene chips. Methods Seven workers who were diagnosed as benzene poisoning (1 suspected, 2 mild, 2 moderate, and 2 severe) were selected and compared with healthy subjects who were matched in age and length of service and were separated from their peripheral blood Total RNA was extracted from mononuclear cells and purified by Trizol kit. The cDNA products were reverse transcribed and labeled with fluorescent dyes Cy3 and Cy5, respectively, to hybridize with 177 microarray chips containing apoptosis-related genes. Results A total of 41 apoptosis-related genes were abnormally expressed. Among them, 3 genes were up-regulated in light and moderate benzene workers. One gene was up-regulated in all benzene poisoning workers, As the degree increased, the total number of differentially expressed genes associated with apoptosis decreased. Conclusion The differential expression of apoptosis related genes in benzene poisoning patients is involved in the occurrence and development of benzene hematological toxicity.
