目的探究Twist介导的衰老逃逸在宫颈癌侵袭转移过程中的作用。方法免疫组织化学法检测70例宫颈鳞状细胞癌标本中Twist和衰老指标CBX3的表达;合成靶向Twist的小干扰RNA瞬时转染宫颈鳞癌细胞SiHa,免疫细胞化学及β-半乳糖苷酶染色检测细胞衰老变化。结果 Twist在早期宫颈鳞癌(Ⅰa~Ⅱa期)及晚期宫颈鳞癌(Ⅱb~Ⅲb期)组织中表达强阳性率分别为6.7%和15.0%,CBX3表达强阳性率分别为10.0%和0%;而在发生或未发生淋巴结转移的宫颈鳞癌组织中,Twist表达强阳性率分别为25.0%和2.4%,CBX3表达强阳性率分别为0%和7.2%。Twist在早、晚期宫颈鳞癌组织表达改变与CBX3相反。低表达Twist的SiHa中CBX3表达上调,并伴有β-半乳糖苷酶染色增强。结论 Twist在宫颈癌发展过程中表达逐渐升高,其介导的衰老逃逸贯穿宫颈癌侵袭转移的整个过程。
Objective To investigate the role of Twist-mediated aging escape in cervical cancer invasion and metastasis. Methods Immunohistochemical method was used to detect the expression of Twist and aging index CBX3 in 70 cervical squamous cell carcinoma specimens. The small interfering RNA targeting Twist was transiently transfected into SiHa cells, immunocytochemistry and β-galactosidase Staining to detect cellular senescence. Results The strong positive rates of Twist expression in early cervical squamous cell carcinoma (Ⅰa ~ Ⅱa) and advanced cervical squamous cell carcinoma (Ⅱb ~ Ⅲb) tissues were 6.7% and 15.0%, respectively. The positive rates of CBX3 were 10.0% and 0% The positive rates of Twist expression in cervical squamous cell carcinoma with or without lymph node metastasis were 25.0% and 2.4%, respectively. The strong positive rates of CBX3 expression were 0% and 7.2% respectively. Twist in early and late cervical squamous cell carcinoma changes in the expression and CBX3 contrary. CBX3 expression was up-regulated in SiHa with low expression of Twist, accompanied by enhanced β-galactosidase staining. Conclusions Twist expression gradually increases during the development of cervical cancer, which is mediated by the aging escape through the entire process of cervical cancer invasion and metastasis.
