目的:研究3天和3周间歇运动对大鼠心肌和冠状血管肾上腺髓质素(Adrenomedullin,ADM)的影响,探讨短期和长期间歇运动对心肌和冠状血管保护作用的机制。方法:25只SD大鼠,参照文献建立短期和长期间歇运动动物模型,以30m/min的速度在跑台上运动15min、休息5min,重复3次。分别以这种方式连续运动3天(3D组,n=12)和3周(3W组,n=13),另取12只大鼠不运动作为对照组(C组)。采用酶联免疫法检测大鼠血清和心肌ADM含量,免疫组织化学法检测心肌和冠状血管ADM表达,并应用计算机图像分析技术对心肌和冠状血管阳性表达面积和平均光密度进行定量分析。结果:与C组比较,3D组和3W组血清ADM含量变化不明显,3D组心肌ADM含量变化不明显,3W组心肌ADM含量明显升高(P<0.05);与C组比较,3D组心肌和冠状血管ADM表达无明显变化,3W组心肌ADM表达显著增强。计算机图像分析结果显示,3W组心肌ADM阳性表达面积和平均光密度明显高于C组(P<0.05),冠状血管阳性表达面积和平均光密度明显低于C组(P<0.05)。结论:长期间歇运动可增加心肌ADM分泌,可能有利于维持血液动力学稳定和保护心肌。
AIM: To investigate the effects of 3 and 3 week intermittent exercises on myocardial and coronary adrenomedullin (ADM) in rats and to explore the mechanism of myocardial and coronary protective effect of short-term and long-term intermittent exercises. Methods: Twenty-five Sprague-Dawley rats were used to establish animal models of short-term and long-term intermittent exercise with reference to the literature. The rats were exercised at a speed of 30 m / min for 15 min on a treadmill and were rested for 5 min for 3 times. The animals were continuously exercised in this manner for 3 days (3D group, n = 12) and 3 weeks (3W group, n = 13), respectively. The content of ADM in serum and myocardium was detected by enzyme-linked immunosorbent assay. The expression of ADM in myocardium and coronary vessels was detected by immunohistochemical method. The area and average optical density of myocardium and coronary vessels were quantified by computer image analysis. Results: Compared with group C, the content of ADM in 3D group and 3W group did not change obviously. The content of ADM in 3D group did not change obviously, and the content of ADM in 3W group was significantly increased (P <0.05). Compared with C group, And coronary artery ADM expression had no significant change, 3W group myocardial ADM expression was significantly enhanced. Computer image analysis showed that the positive expression area and average optical density of myocardium in 3W group were significantly higher than those in C group (P <0.05). The positive expression area and average optical density of coronary vessels in group 3W were significantly lower than those in group C (P <0.05). CONCLUSIONS: Long-term intermittent exercise increases myocardial ADM secretion, which may be beneficial to maintain hemodynamic stability and protect myocardium.
