Objective: To investigate the effect of estrogen on spontaneous depression after cerebral ischemia in rats and its effect on the expression of brain-derived neurotrophic factor (BDNF) in penumbra of ischemic penumbra. Methods: The cerebral ischemia model was established by middle cerebral artery occlusion. Rats with cerebral ischemia were randomly divided into control group, stroke group and estrogen intervention group (subcutaneous injection of 17 mL 17-estradiol soybean oil 0.1 mL for 2 weeks). Depression behavioral changes after cerebral ischemia and estrogen intervention were observed to detect BDNF expression in ischemic penumbra. Results: Rats were forced to swim for an extended period of time after cerebral ischemia. In the estrogen group, the immobility time was shortened and the number of BDNF positive cells in the penumbra was significantly higher than that in the stroke group. CONCLUSION: Depression-like symptoms can spontaneously develop after cerebral ischemia in rats. Estrogen treatment may improve these symptoms and improve the expression of BDNF in ischemic penumbra may be one of its therapeutic mechanisms.
