目的 :探讨水蛭素对体外培养的巩膜成纤维细胞及细胞外基质的影响。方法 :用体外培养巩膜成纤维细胞第 3代 ,将不同浓度的水蛭素 5 0U/ml、4 0U/ml、2 0U/ml、10U/ml、1U/ml分别加入等体积的培养液 2 0 0 μl,每 7孔为一浓度组 ,无药培养液为对照组 ,共 6组 ,培养 4 8小时后用MTT法测定各孔贴壁细胞的吸光值 (A) ,取其上清液用放射免疫法测定Ⅲ型胶原及HA的量。结果 :水蛭素 10U/ml组A值与对照组相比有极显著差异 (P <0 0 1) ,其增生抑制率最高。水蛭素 10U/m组HA、Ⅲ型胶原的浓度明显低于对照组 ,有极显著性差异 (P <0 0 1) ;其余几组中HA的浓度与对照组相比有显著差异 (P <0 0 5 )。结论 :水蛭素对体外培养巩膜成纤维细胞及细胞外基质有抑制作用
Objective: To investigate the effect of hirudin on cultured scleral fibroblasts and extracellular matrix. Methods: The third generation of scleral fibroblasts were cultured in vitro, and different concentrations of hirudin 50U / ml, 40U / ml, 20U / ml, 10U / ml and 1U / ml were added into the same volume of culture medium 20 0 μl, a concentration of 7 holes for each group, drug-free culture medium for the control group, a total of 6 groups, cultured 48 hours after the MTT assay for each well adherent cells absorbance (A), the supernatant Radioimmunoassay was used to determine the amount of type Ⅲ collagen and HA. Results: The value of A in hirudin 10U / ml group was significantly different from that in control group (P <0.01), and the inhibition rate of hirudin was the highest. The concentrations of HA and type Ⅲ collagen in hirudin 10U / m group were significantly lower than those in control group (P <0.01). The HA concentration in hirudin group was significantly lower than that in control group (P < 0 0 5). Conclusion: Hirudin can inhibit the growth of scleral fibroblasts and extracellular matrix in vitro
