[目的]探讨了检测外周血低氧诱导因子-1α(HIF-1α)和胰岛素样生长因子-I(IGF-1)因子表达变化在溃疡性结肠炎(UC)患者中的临床意义。[方法]选取2014年1月~2016年4月我院诊治的UC患者76例,另入选40例健康体检者为正常对照。HIF-1α和IGF-1因子表达的检测使用酶联免疫吸附法(ELISA)。[结果]UC患者HIF-1α和IGF-1表达分别为(38.64±7.53)ng/ml和(57.82±12.26)ng/ml,均显著高于健康对照组的(12.75±3.16)ng/ml和(26.35±5.14)ng/ml(P<0.01)。活动期UC患者HIF-1α和IGF-1表达分别为(44.52±8.37)ng/ml和(64.65±13.14)ng/ml,均显著高于缓解期UC患者的(19.68±4.34)ng/ml和(5.73±6.58)ng/ml(P<0.01)。不同临床严重程度UC患者间HIF-1α和IGF-1表达差异有统计学意义(P<0.05)。其中重型患者HIF-1α和IGF-1表达显著高于中型患者(P<0.05),中型患者HIF-1α和IGF-1表达又显著高于轻型患者(P<0.05)。UC患者不同内镜分级间HIF-1α和IGF-1表达差异有统计学意义(P<0.05)。其中Ⅲ级患者HIF-1α和IGF-1表达显著高于Ⅱ级患者(P<0.05),Ⅱ级患者HIF-1α和IGF-1表达又显著高于Ⅰ级患者(P<0.05)。UC患者HIF-1α和IGF-1表达呈显著正相关(r=0.517,P<0.05)。[结论]UC患者存在HIF-1α和IGF-1异常高表达,HIF-1α和IGF-1的检测有可能用于UC患者病情的评估。
[Objective] To explore the clinical significance of detecting the expression of hypoxia-inducible factor-1α (HIF-1α) and insulin-like growth factor-I (IGF-1) in peripheral blood of patients with ulcerative colitis (UC). [Methods] Totally 76 UC patients diagnosed and treated in our hospital from January 2014 to April 2016 were enrolled, and 40 healthy subjects were selected as normal control. The detection of HIF-1α and IGF-1 factor expression was detected by enzyme-linked immunosorbent assay (ELISA). [Results] The expressions of HIF-1α and IGF-1 in UC patients were (38.64 ± 7.53) ng / ml and (57.82 ± 12.26) ng / ml respectively, which were significantly higher than those in healthy controls (12.75 ± 3.16 ng / ml and (26.35 ± 5.14) ng / ml (P <0.01). The expressions of HIF-1α and IGF-1 in active UC patients were (44.52 ± 8.37) ng / ml and (64.65 ± 13.14) ng / ml respectively, which were significantly higher than those in patients with remission UC (19.68 ± 4.34 ng / ml and (5.73 ± 6.58) ng / ml (P <0.01). There were significant differences in the expression of HIF-1α and IGF-1 between UC patients with different clinical severity (P <0.05). The expression of HIF-1α and IGF-1 in severe patients was significantly higher than that in mid-sized patients (P <0.05). The expression of HIF-1α and IGF-1 was also significantly higher in moderate patients than in light patients (P <0.05). There was significant difference in the expression of HIF-1αand IGF-1 among different endoscopic grades of UC patients (P <0.05). The expression of HIF-1α and IGF-1 in grade Ⅲ patients were significantly higher than those in grade Ⅱ patients (P <0.05). The expression of HIF-1α and IGF-1 in grade Ⅱ patients were significantly higher than those in grade Ⅰ patients (P <0.05). There was a significant positive correlation between the expression of HIF-1α and IGF-1 in UC patients (r = 0.517, P <0.05). [Conclusion] The abnormal expression of HIF-1α and IGF-1 exists in UC patients. The detection of HIF-1α and IGF-1 may be used for the evaluation of UC patients.
