目的分析基质金属蛋白酶2(MMP-2)和MMP-9及其抑制因子1(TIMP-1)和TIMP-2在宫颈癌不同位置中的表达情况及其临床意义。方法选取经病理证实的宫颈浸润癌患者118例(ICC组)、宫颈上皮内瘤样病变患者75例(CIN组),取病变中心组织和边缘组织;选取正常宫颈组织标本45例为对照组。采用SABC法行免疫组化检测各组中MMP-2、MMP-9、TIMP-1和TIMP-2因子表达阳性率、染色强度和表达强度。比较各组相关因子表达情况差异。结果 ICC组中MMP-2和MMP-9的阳性率、染色强度和表达强度显著高于CIN组和对照组,TIMP-1和TIMP-2的阳性率、染色强度和表达强度显著低于CIN组和对照组(P<0.05)。在ICC组,边缘癌组织的MMP-2与MMP-9的阳性率、染色强度和表达强度明显高于中心癌组织,而TIMP-1与TIMP-2的阳性率、染色强度和表达强度明显低于中心癌组织(P<0.05)。结论 MMP-2、MMP-9及其抑制因子TIMP-1、TIMP-2与宫颈癌的发生、发展密切相关,可能在宫颈癌的侵袭与转移中发挥着重要作用。
Objective To analyze the expression of MMP-2, MMP-9, TIMP-1 and TIMP-2 in different sites of cervical cancer and its clinical significance. Methods One hundred and eighteen patients (ICC group) and 75 patients with cervical intraepithelial neoplasia (CIN group) were enrolled in the study. 45 cases of normal cervical tissue specimens were selected as the control group. The positive rate, staining intensity and expression intensity of MMP-2, MMP-9, TIMP-1 and TIMP-2 in each group were detected by SABC immunohistochemical method. Compare the expression of related factors in each group. Results The positive rate, staining intensity and intensity of MMP-2 and MMP-9 in ICC group were significantly higher than those in CIN group and control group. The positive rates of TIMP-1 and TIMP-2 in ICC group were significantly lower than those in CIN group And control group (P <0.05). In the ICC group, the positive rates of MMP-2 and MMP-9 in the marginal cancer tissues were significantly higher than those in the central cancer tissues, while the positive rates of TIMP-1 and TIMP-2, the staining intensity and the expression intensity were significantly lower In central carcinoma (P <0.05). Conclusion The expressions of MMP-2, MMP-9, TIMP-1 and TIMP-2 are closely related to the occurrence and development of cervical cancer, which may play an important role in the invasion and metastasis of cervical cancer.
