目的:探讨2’-羟基-4’,6’-二异戊烯氧基查尔酮对肺癌A549细胞的增殖抑制作用和对细胞凋亡的影响;方法:利用MTT法检测2’-羟基-4’,6’-二异戊烯氧基查尔酮对肺癌A549细胞增殖抑制作用,并测定其IC_(50);利用Hochest染色观察2’-羟基-4’,6’-二异戊烯氧基查尔酮对肺癌A549细胞凋亡的形态学变化;流式细胞术检测2’-羟基-4’,6’-二异戊烯氧基查尔酮对肺癌A549细胞早期凋亡率的影响;结果:与空白对照组比较,各剂量组(5、10、20μmol/L)2’-羟基-4’,6’-二异戊烯氧基查尔酮能有效抑制肺癌A549肺癌细胞增殖(P<0.05),并可显著诱导A549细胞发生早期凋亡(P<0.05),其作用呈时间浓度依赖性。结论:2’-羟基-4’,6’-二异戊烯氧基查尔酮能有效抑制肺癌A549细胞的增殖,这种增殖抑制作用可能与2’-羟基-4’,6’-二异戊烯氧基查尔酮能够诱导A549细胞发生凋亡有关。
AIM: To investigate the inhibitory effect of 2’-hydroxy-4 ’, 6’-diisopentenyloxychalcone on the proliferation of lung cancer A549 cells and its effect on apoptosis.Methods: MTT assay was used to detect the 2’-hydroxy- The inhibitory effect of 4 ’, 6’-diisopentenyloxychalcone on the proliferation of lung cancer A549 cells was determined and its IC 50 was determined. The effect was time-dependent. CONCLUSION: 2’-hydroxy-4 ’, 6’-diisopentenyloxy chalcone can effectively inhibit the proliferation of lung cancer A549 cells. The inhibitory effect of 2’-hydroxy-4’, 6’- Isopentenoxy Chalcone can induce the apoptosis of A549 cells.
