目的 探讨大鼠中胚层来源的骨髓间充质干细胞(MSCs),在诱导因子的诱导下体外向神经元方向分化的能力。方法 贴壁法分离的MSCs,用NIM诱导,相差显微镜观察细胞形态变化,神经元特异抗体NeuN,MAP2,NSE免疫组化染色鉴定转化情况。结果 在诱导后30~40min即开始形态变化,形成神经元样的细胞,免疫组化染色神经元样细胞表达NeuN(50.83%±3.43%),NSE(59.83%±9.24%)和MAP2(45.17%±8.42%)。结论 MSCs在体外诱导下可以分化为神经元样的细胞,表明它是有别于一般成体干细胞的多能干细胞。
Objective To investigate the ability of rat mesodermal derived bone marrow mesenchymal stem cells (MSCs) to differentiate into neurons in vitro under the induction of inducing factors. Methods The MSCs isolated by adherence method were induced by NIM, the cell morphological changes were observed by phase contrast microscopy, and the neuronal specific antibodies NeuN, MAP2 and NSE were used to identify the transformation. The results showed that neuronal cells were formed 30 ~ 40min after induction, and NeuN (50.83% ± 3.43%), NSE (59.83% ± 9.24%) and MAP2 (45.17% ± 8.42%). Conclusion MSCs can differentiate into neuron-like cells under in vitro induction, indicating that they are pluripotent stem cells that are distinct from the normal adult stem cells.
