目的:建立用高效液相色谱法测定三黄散中黄芩苷和盐酸小檗碱含量的方法。方法:采用Agilent-C18色谱柱(250mm×4.6 mm,5μm),流动相为乙腈-0.02 mol·L-1磷酸二氢钾溶液-三乙胺(25∶75∶0.2)(用磷酸调pH至3.0),柱温为25℃,流速为1.0 ml·min-1,检测波长为333 nm。结果:黄芩苷与盐酸小檗碱分别在0.080 8~0.606 3μg和0.064 7~0.485 1μg范围内线性关系良好,方法平均回收率分别为98.7%和99.6%,RSD分别为0.3%和0.4%(n=9)。结论:该法简便易行,结果可靠准确,两组分分离良好。
Objective: To establish a method for the determination of baicalin and berberine hydrochloride in Sanhuang powder by high performance liquid chromatography. Methods: Agilent-C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase was acetonitrile-0.02 mol·L-1 potassium dihydrogen phosphate solution-triethylamine (25:75:0.2) 3.0). The column temperature was 25 ℃, the flow rate was 1.0 ml · min-1 and the detection wavelength was 333 nm. Results: The linear relationship was found between baicalin and berberine in the range of 0.080 8 ~ 0.606 3μg and 0.064 7 ~ 0.485 1μg respectively. The average recoveries were 98.7% and 99.6%, respectively. The RSDs were 0.3% and 0.4% respectively = 9). Conclusion: The method is simple and easy to operate, the results are reliable and accurate, and the two components are well separated.
