目的 本实验观察阿魏酸钠对炎症介质组胺引起的血管内皮通透性增加的作用。方法 从新生牛主动脉分离获得单个内皮细胞 ,培养于微孔滤膜上直至形成致密单层。通过灌流Hanks液或含 5g·L-1白蛋白的Hanks液后 ,测定经 1× 10 -4 mol·L-1组胺溶液加 1× 10 -5mol·L-1阿魏酸钠处理的液体滤过系数 (Kf)、液体滤过流量 (Jv)和蛋白质渗透压反射系数 (σ)。结果 内皮单层经组胺处理后 ,Kf和Jv降低 ,σ升高 ;而阿魏酸钠能够抑制因组胺造成的Kf和Jv降低 ,以及σ升高。结论 组胺能够引起血管内皮单层通透性的增加 ,而阿魏酸钠能够减轻组胺造成的内皮单层通透性的增加 ;其作用机制需要进一步地研究。
Objectives To observe the effect of sodium ferulate on the increase of histamine-induced vascular endothelial permeability in the inflammatory mediators. Methods A single endothelial cell was isolated from the bovine aorta and cultured on a microporous membrane until a dense monolayer was formed. After perfusion of Hanks solution or Hanks solution containing 5 g·L-1 albumin, the liquid treated with 1×10 -4 mol·L -1 histamine solution plus 1 × 10 -5 mol·L -1 sodium ferulate was determined. The filtration coefficient (Kf), the liquid filtration flux (Jv) and the protein osmotic pressure reflection coefficient (σ). Results After the histamine treatment of the endothelial monolayer, the Kf and Jv decreased and the σ increased, while the sodium ferulate inhibited the decrease of Kf and Jv caused by histamine and the increase of σ. Conclusion Histamine can increase the permeability of vascular endothelial monolayer, and sodium ferulate can reduce the increase of histamine-induced endothelial monolayer permeability. Its mechanism of action needs further study.
