目的研究铅对乳鼠成骨细胞凋亡相关基因表达的影响。方法分离并培养原代成骨细胞,加入不同浓度(0、1、10、100μmol/L)Pb(Ac)2,培养72h,用免疫组化染色法检测成骨细胞凋亡相关基因表达的情况。结果铅处理组成骨细胞Fas基因表达增加(r=0·78),而Bcl-2基因表达减少(r=-0·82)。结论铅对成骨细胞的影响可能与Fas和Bcl-2基因表达改变有关。
Objective To study the effect of lead on the expression of apoptosis related genes in cultured neonatal rat osteoblasts. Methods Primary osteoblasts were isolated and cultured. Pb (Ac) 2 with different concentrations (0, 1, 10 and 100μmol / L) was added and cultured for 72h. The expression of apoptosis related genes in osteoblasts was detected by immunohistochemistry . Results Lead treatment increased the expression of Fas gene (r = 0.78) and decreased the expression of Bcl-2 gene (r = -0.82). Conclusion The effect of lead on osteoblasts may be related to the changes of Fas and Bcl-2 gene expression.
