在评估抗人活化血小板α-颗粒膜蛋白(GMP-140)单克隆抗体(McAb)SZ-51与体内血栓结合能力的基础上,用~(99m)Tc标记单抗SZ-51对犬颈内动脉血栓模型进行免疫显像。结果表明,单抗SZ-51在循环状态下与血栓有较强的结合能力;~(99m)Tc-SZ-51能清晰地显示实验性犬颈内动脉血栓的部位和范围,血栓最佳显示时间为注射~(99m)Tc-SZ-51后2~4h。注射~(99m)Tc-SZ-51后6~8h处死动物,离体血栓/血液放射活性比值(±S)为6.03±1.09。~(99m)Tc-SZ-51具有活体内导向定位血栓的能力,用于血栓性疾病的放免显像诊断有一定的可行性。
On the basis of evaluating the binding ability of SZ-51 with anti-human activated platelet α-granule membrane protein (GMP-140) monoclonal antibody (McAb) SJ-51 to thrombosis in vivo, Arterial thrombosis model for immunosurgery. The results showed that the monoclonal antibody SZ-51 had strong binding ability to thrombus in the circulatory state; ~ (99m) Tc-SZ-51 could clearly show the location and range of the experimental canine internal carotid artery thrombosis and the optimal thrombus Time after injection ~ (99m) Tc-SZ-51 2 ~ 4h. After 6 ~ 8h injection of ~ (99m) Tc-SZ-51, the in vitro thrombus / blood radioactivity ratio (± S) was 6.03 ± 1.09. ~ (99m) Tc-SZ-51 has the ability of targeting thrombosis in vivo and has some feasibility for the diagnosis of thromboembolic disease.
