目的观察和分析TGF-β1对子宫内膜基质细胞(endometrial stromal cells,ESCs)增殖和活性的影响。方法体外分离、培养和鉴定ESCs。实验分组:对照组和TGF-β1组,对照组:加入不含血清的培养液;转化生长因子-β1(TGF-β1)组:不含血清的培养液+不同浓度的TGF-β1,通过噻唑蓝法检测各组细胞的增殖率;用ELISA法检测各组细胞培养液中Ⅰ型胶原(Collagen typeⅠ,ColⅠ)和纤黏连蛋白(Fibronectin,FN)的含量。免疫印迹法检测各组细胞胞质中的MMP-9及TIMP-1的表达水平。结果 TGF-β1可以促进ESCs增殖,并呈剂量依赖性;使细胞分泌ColⅠ(0.736 6±0.042 4)和FN(0.713 1±0.046 8)的量增多,细胞内MMP-9含量减低,而TIMP-1含量升高;与对照组比较,其差异有统计学意义(P<0.05)。结论 TGF-β1能使子宫内膜基质细胞增殖,并使其活性增加。
Objective To observe and analyze the effect of TGF-β1 on proliferation and activity of endometrial stromal cells (ESCs). Methods The ESCs were isolated, cultured and identified in vitro. The experimental groups were as follows: control group and TGF-β1 group, control group: adding serum-free medium; TGF-β1 group: serum-free culture medium + different concentrations of TGF-β1, The proliferation rate of cells in each group was detected by the blue method. The content of collagen typeⅠ (ColⅠ) and fibronectin (FN) in each group were detected by ELISA. Western blotting was used to detect the expression of MMP-9 and TIMP-1 in the cytoplasm of each group. Results TGF-β1 promoted the proliferation of ESCs in a dose-dependent manner. The amount of ColⅠ (0.736 ± 0.042 4) and FN (0.713 1 ± 0.046 8) secreted by TGF-β1 increased and the content of MMP- 1 content increased; compared with the control group, the difference was statistically significant (P <0.05). Conclusion TGF-β1 can proliferate endometrial stromal cells and increase their activity.
