目的观察降糖补肾方对小剂量链脲佐菌素(STZ)加高脂饲料诱导的糖尿病模型大鼠肌肉组织IKK激酶β(IKKβ)信使核糖核酸(mRNA)表达的影响。方法雄性SD大鼠高脂饲料喂养4周后注射小剂量STZ,1周后测大鼠空腹血糖,血糖值>11.1 mmol/L者为造模成功的大鼠,将造模成功的大鼠随机分为模型组和降糖补肾方干预组,另设阿司匹林对照组和空白对照组,药物干预4周,取大腿肌肉组织,采用聚合酶链反应逆转录RT-PCR技术检测IKKβmRNA表达水平。结果模型组IKKβmRNA表达明显高于空白对照组,是空白对照组的5.098倍(P<0.01);阿司匹林组、降糖补肾高、中剂量组能明显降低IKKβmRNA表达,分别是空白对照组的2.814、3.246、3.770倍,与模型组比P<0.01;降糖补肾低剂量组能降低IKKβmRNA表达,是空白对照组的4.974倍,与模型组比P>0.05。结论降糖补肾方在一定程度上能抑制糖尿病大鼠IKKβmRNA基因,这可能是降糖补肾方抑制炎症信号通路的传导而发挥其改善胰岛素抵抗治疗糖尿病的机制之一。
Objective To observe the effect of Jiangtang Bushen Recipe on the mRNA expression of IKKβ in muscle tissue of streptozotocin (STZ) plus high lipid diet-induced diabetic rats. Methods Male SD rats were fed with high-fat diet for 4 weeks and then injected with low-dose STZ. Fasting blood glucose was measured after 1 week and blood glucose> 11.1 mmol / L was used as successful model rats. The successful rats were randomized The rats in model group and Jiangtang Bushen Fang intervention group were divided into two groups: aspirin control group and blank control group. Drug intervention for 4 weeks, thigh muscle tissue was taken and the expression of IKKβ mRNA was detected by polymerase chain reaction reverse transcription-polymerase chain reaction (RT-PCR). Results The expression of IKKβmRNA in model group was significantly higher than that in control group (5.098-fold) (P <0.01). Aspirin group, high and middle dosage of Jiangtang Bushen group could significantly reduce IKKβmRNA expression, which were respectively 2.814, 3.246,3.770 folds, which was significantly lower than that of the model group (P <0.01). The low-dose HGT low dose group could reduce the expression of IKKβmRNA, which was 4.974 times of the blank control group, P <0.05 compared with the model group. Conclusion Jiangtang Bushen Prescription can inhibit IKKβ mRNA in diabetic rats to some extent, which may be one of the mechanisms that Jiangtangshenzheng Prescription can inhibit the transmission of inflammatory signal transduction and play its role in improving diabetes mellitus.
