目的探讨复方血栓通胶囊对糖尿病大鼠肾脏的保护作用。方法雄性SD大鼠予链脲佐菌素(STZ)60mg/kg单次腹腔注射诱发糖尿病,3d后尾静脉血糖高于16.7mmol/L者视为造模成功。实验动物分为正常对照组(NG)、糖尿病组(DM)、糖尿病复方血栓通胶囊处理组(XST)、糖尿病α-硫辛酸处理组(ALA),每组均10只。12周后比较各组体质量、血糖、肾脏/体质量、血脂、24h尿蛋白含量、血肌酐、尿素氮。透射电镜及光镜观察肾脏超微结构改变。结果与正常对照组相比,DM组大鼠24h尿蛋白排泄量明显增加(P<0.05),经复方血栓通及α-硫辛酸处理后,24h尿白蛋白排泄量明显降低(P<0.05);与正常对照组相比,各模型组大鼠左肾质量/体质量比明显升高,差异有统计学意义(P<0.01),经复方血栓通胶囊、α-硫辛酸处理后大鼠左肾质量/体质量比降低,其中XST组与DM组相比差异有统计学意义(P<0.05)。电镜下可见DM组大鼠肾小球基底膜厚度不均匀增厚,足突融合、消失,系膜细胞增生;经复方血栓通胶囊、α-硫辛酸处理后,上述病变有所好转。结论复方血栓通胶囊可降低24h尿白蛋白排泄量,降低肾脏肥大系数(左肾质量/体质量),改善肾小球基底膜结构,对糖尿病大鼠肾脏具有保护作用。
Objective To investigate the protective effect of compound Xueshantong capsule on the kidney of diabetic rats. Methods Male Sprague-Dawley rats were given intraperitoneal injection of streptozotocin (STZ) 60mg / kg to induce diabetes mellitus, and the tail vein glucose was higher than 16.7mmol / L after 3 days. The experimental animals were divided into normal control group (DM), diabetic group (XST), diabetic α-lipoic acid treatment group (ALA), 10 in each group. After 12 weeks, body weight, blood glucose, kidney / body weight, blood lipid, proteinuria in 24 hours, serum creatinine and urea nitrogen were compared. The changes of renal ultrastructure were observed by transmission electron microscope and light microscope. Results Compared with the normal control group, the excretion of 24 h urinary protein in DM group was significantly increased (P <0.05). After excretion of Xueshuantong and α-lipoic acid, the urinary albumin excretion decreased significantly (P <0.05) ; Compared with the normal control group, the left kidney mass / body mass ratio of each model group was significantly increased, the difference was statistically significant (P <0.01), the compound Xuesaitong capsule, α-lipoic acid treatment of rats left Kidney mass / body mass ratio decreased, among which the difference between XST group and DM group was statistically significant (P <0.05). Electron microscope shows that DM group rats glomerular basement membrane thickness unevenly thick, foot processes fusion, disappearance, mesangial cell proliferation; the compound Xueshuantong capsule, α-lipoic acid treatment, the lesions improved. Conclusion Compound Xueshuantong Capsule can reduce 24 h urinary albumin excretion, reduce renal hypertrophy (left kidney mass / body weight), improve glomerular basement membrane structure, and protect kidney of diabetic rats.
