目的探讨新型棉酚衍生物apogossypol对胃癌SGC7901移植瘤抑制及免疫功能之间的关系。方法体外培养扩增SGC7901胃癌细胞,接种裸鼠,建立小鼠移植瘤模型。计算肿瘤的抑瘤率,测定自然杀伤(NK)细胞活性,腹腔巨噬细胞一氧化氮(NO)的产生量,NK细胞活性,T淋巴细胞增殖能力及白细胞介素-2(IL-2)的活性。结果 Apogossypol可明显提高荷瘤小鼠吞噬细胞的功能并抑制肿瘤的增殖,NO产生量增加并可显著促进IL-2分泌,进而影响淋巴细胞的活性,最终导致NK细胞功能增强。结论 Apogossypol可增强荷瘤小鼠的免疫功能,抑制肿瘤的生长。
Objective To investigate the relationship between apogossypol, a new gossypol derivative, on the inhibition and immune function of gastric cancer SGC7901 xenografts. Methods SGC7901 gastric cancer cells were cultured and expanded in vitro. Nude mice were inoculated to establish a transplanted tumor model in mice. The tumor inhibition rate was calculated. The activity of natural killer (NK) cells, the production of nitric oxide (NO) in peritoneal macrophages, the activity of NK cells, the proliferation of T lymphocytes and the activity of interleukin-2 (IL- Activity. Results Apogossypol significantly increased phagocytic function of tumor-bearing mice and inhibited the proliferation of tumor cells. The increased amount of NO produced a significant increase of IL-2 secretion and then the activity of lymphocytes, leading to enhanced NK cell function. Conclusion Apogossypol can enhance the immune function of tumor-bearing mice and inhibit tumor growth.
