目的:总结丙酮酸激酶M2型(PKM2)在肿瘤诊断与治疗中的应用的研究进展。方法:应用PubMed检索系统,以“PKM2”为关键词,检索2001-01-2013-02相关文献。纳入标准:1)PKM2功能与调节;2)PKM2在肿瘤诊断中的应用;3)PKM2在肿瘤治疗中的应用。根据纳入标准,符合分析的文献45篇。结果:PKM2在肿瘤中特异表达,并在肿瘤代谢中起着重要的作用。在多种类型的肿瘤患者血液和粪便中检测到PKM2含量升高,并与肿瘤分化、转移与分期相关,在肿瘤诊断与监测中有着重大的价值。促进或抑制PKM2都抑制肿瘤的生长。目前,已有多种针对PKM2的药物和小分子化合物的研究,并且改变PKM活性对多种化疗药物有着增敏的作用。结论:PKM2作为一种新型肿瘤标志有着良好的应用前景,而对PKM2干预为肿瘤的治疗提供了新的思路。
Objective: To summarize the research progress of pyruvate kinase M2 (PKM2) in the diagnosis and treatment of tumors. Methods: PubMed retrieval system was used, and the key words “PKM2” were retrieved from 2001-01-2013-02. Inclusion criteria: 1) PKM2 function and regulation; 2) PKM2 in the diagnosis of cancer; 3) PKM2 in the treatment of cancer. According to the inclusion criteria, 45 articles were analyzed. RESULTS: PKM2 was specifically expressed in tumors and plays an important role in tumor metabolism. Detection of elevated levels of PKM2 in blood and faeces of many types of oncology patients is associated with tumor differentiation, metastasis and staging, and is of great value in the diagnosis and monitoring of tumors. Promoting or inhibiting PKM2 all inhibit tumor growth. Currently, there are a variety of drugs and small molecule compounds for PKM2 research, and to change PKM activity on a variety of chemotherapeutic drugs have sensitization effect. Conclusion: PKM2 has a good prospect of application as a new tumor marker, and provides a new idea for the treatment of tumor with PKM2 intervention.
