目的:建立平菇多糖的含量测定方法,确定纤维素酶法提取平菇多糖的最佳工艺条件。方法:采用苯酚-硫酸法测定平菇多糖的含量。以平菇多糖得率为指标,考察酶解时间、酶解温度、pH值等3个因素对平菇多糖纤维素酶提取工艺的影响,采用响应面分析法优化工艺条件。结果:平菇多糖的最佳提取工艺条件为酶解时间135min、酶解温度49℃、pH5.30,在此条件下得率可达6.82%。结论该法成本低、产品得率高,比传统水提法提高3.98%。
Objective: To establish a method for the determination of polysaccharides in Pleurotus ostreatus, and to determine the optimal conditions for extracting polysaccharides from Pleurotus ostreatus by cellulase. Methods: The polysaccharide content of Pleurotus ostreatus was determined by phenol - sulfuric acid method. Taking polysaccharide yield of Pleurotus ostreatus as index, the effects of enzymolysis time, enzymolysis temperature and pH value on the extraction of polysaccharides from Pleurotus ostreatus were investigated. The reaction conditions were optimized by response surface analysis. Results: The optimal extraction conditions of Pleurotus ostreatus polysaccharides were enzymolysis time 135min, enzymolysis temperature 49 ℃, pH5.30, the yield of polysaccharides reached 6.82%. Conclusion The method has low cost and high product yield, which is 3.98% higher than the traditional water extraction method.
