目的:观察人肺癌裸鼠移植瘤整合蛋白表达与其转移的相关性。方法:以ABC免疫组化染色,对转移力由低到高依次为PLA-801C、Anip973、PLA-801D、PLA-801DL四株人肺癌裸鼠移植瘤作整合蛋白亚单位表达的观察,并在裸鼠体内观察整合蛋白β1抗体及其配体RGD多肽对移植瘤生长及转移的影响。结果:四株移植瘤均呈α2、α5、α6B阴性。β1的表达除Anip973呈强阳性、PLA-801DL呈弱阳性外,也均呈阴性。但四株移植瘤除PLA-801C外,其余3株均呈α6A强阳性。β1抗体及RGD多肽对PLA-801DL移植瘤的转移不见明显影响。结论:4株人肺癌移植瘤总体上呈β1亚族整合蛋白低表达,表达水平与其转移无显著相关。
Objective: To observe the correlation between the expression of integrin and its metastasis in human lung cancer xenografts in nude mice. Methods: Immunohistochemical staining with ABC was used to observe the expression of integrin subunits in lung cancer xenografts from PLA-801C, Anip973, PLA-801D and PLA-801DL. The effects of integrin β1 antibody and its ligand RGD polypeptide on growth and metastasis of transplanted tumors were observed in nude mice. Results: All four tumors were negative for α2, α5 and α6B. The expression of β1 was negative except that Anip973 was strongly positive and PLA-801DL was weakly positive. However, except for PLA-801C, all the other 4 tumors showed strong α6A positive. Β1 antibody and RGD polypeptide had no significant effect on the migration of PLA-801DL xenografts. CONCLUSIONS: Four human lung cancer xenografts exhibit low expression of the integrin β1 subfamily, and their expression levels are not significantly associated with their metastasis.
