【目的】探索离体条件下铁皮石斛体细胞胚发生过程,为铁皮石斛的快速繁殖生产和种质资源保存提供技术依据。【方法】取继代培养30 d的铁皮石斛类原球茎作接种材料,以N6为基本培养基,加入植物激素,并以真菌提取物作诱导子,对类原球茎和愈伤组织进行诱导培养。【结果】显微观察结果表明铁皮石斛体细胞胚的形成可由愈伤组织的表皮细胞或内部细胞产生。【结论】铁皮石斛通过类原球茎和愈伤组织的诱导培养能获得大量的丛芽。
【Objective】To explore the process of somatic embryogenesis of Dendrobium candidum under in vitro conditions and provide technical basis for rapid propagation of Dendrobium candidum and preservation of germplasm resources. 【Method】The seedlings of Dendrobium candidum were used as inoculum for 30 days, and N6 was used as the basic medium. Phytohormone was added. The fungal extracts were used as inducers to induce the protocorm-like bodies and callus. . [Results] Microscopic observations revealed that the formation of somatic embryos of Dendrobium candidum can be produced by the epidermal cells or internal cells of the callus. 【Conclusion】 A large number of cluster buds can be obtained from the induction culture of D. candidum by protocorm-like and callus.
