目的:探讨乙酰肝素酶(heparanase,HPA)及缺氧诱导因子-1α(hypoxia inducible factors-1α,HIF-1α)在食管鳞癌中的表达及其与食管鳞癌浸润转移的关系。方法:应用原位杂交技术检测70例食管鳞癌组织和23例切缘正常组织中HPA mRNA表达,同时应用免疫组化法检测HIF-1α蛋白在全部标本中的表达。结果:癌组织中HPA mRNA和HIF-1α的阳性表达率分别为65.7%(46/70)和55.7%(39/70),显著高于切缘正常组织(P<0.05);HPA mRNA和HIF-1α的表达与食管鳞癌组织浸润深度、淋巴结转移有关(P<0.05);在食管鳞癌组织中HPA mRNA的表达与HIF-1α的表达关系密切(Kappa=0.615,P<0.05)。结论:HPA和HIF-1α阳性表达与食管鳞癌的生长及浸润转移有关,两者在食管鳞癌的发生、发展过程中可能存在着一定的协同作用;检测HPA与HIF-1α表达有助于食管鳞癌生物学性能的判断。
Objective: To investigate the expression of heparanase (HPA) and hypoxia inducible factors-1α (HIF-1α) in esophageal squamous cell carcinoma and its relationship with invasion and metastasis of esophageal squamous cell carcinoma. Methods: The expression of HPA mRNA in 70 cases of esophageal squamous cell carcinoma and 23 cases of normal marginal tissue was detected by in situ hybridization. At the same time, the expression of HIF-1α protein in all samples was detected by immunohistochemistry. Results: The positive rates of HPA mRNA and HIF-1α in cancerous tissues were 65.7% (46/70) and 55.7% (39/70), respectively, which were significantly higher than those in normal tissues (P <0.05) (P <0.05). The expression of HIF-1α in esophageal squamous cell carcinoma tissues was closely related to the expression of HIF-1α (Kappa = 0.615, P <0.05). Conclusions: The positive expression of HPA and HIF-1α is related to the growth, invasion and metastasis of esophageal squamous cell carcinoma, which may play a synergistic role in the development and progression of esophageal squamous cell carcinoma. Detecting the expression of HPA and HIF-1α is helpful to Biological evaluation of esophageal squamous cell carcinoma.
