为探讨利血平致抑郁及阿米替林抗抑郁的机理,将Wistar雄性大鼠随机分为对照组、利血平模型组和利血平加阿米替林组,测定了大鼠脑内游离钙离子、钙调素、环核苷酸含量变化。结果显示,模型大鼠脑内游离钙离子、钙调素和环核苷酸含量降低,阿米替林可使模型大鼠脑内游离钙离子、钙调素浓度升高,而对环核苷酸则无影响。提示利血平致抑郁的机理与2条第二信使通路均有关,而阿米替林则可能是通过作用于钙信号系统发挥其抗抑郁作用。
In order to explore the mechanism of reserpine-induced depression and amitriptyline antidepressant, Wistar male rats were randomly divided into control group, reserpine model group and reserpine-amitriptyline group. Free calcium, calmodulin, cyclic nucleotide content changes. The results showed that the content of free calcium, calmodulin and cyclic nucleotide in the model rat brain decreased. Amitriptyline increased the concentration of free calcium and calmodulin in the brain of model rats, Acid has no effect. Suggesting that the mechanism of reserpine-induced depression is related to two second messenger pathways, whereas amitriptyline may exert its antidepressant effect by acting on the calcium signaling system.
