目的研究正常子宫内膜、子宫内膜增殖症及子宫内膜癌组织中PTEN、β-CAT蛋白的表达,探讨其与子宫内膜癌发生发展的关系及作为早期癌变生物学指标的可能性。方法应用免疫组化S-P法对26例正常子宫内膜组织、例子55宫内膜增殖症、例子宫内膜癌组织中PTEN、β-CAT蛋白的表达进行研究。结果在正常子宫内膜、子宫内膜增殖症、子宫38内膜腺癌组织中PTEN蛋白的阳性表达率依次递减,差异有显著统计学意义(P<0.01);β-CAT蛋白的阳性表达率依次递增,差异有显著统计学意义(P<0.01)。不同肌层PTEN蛋白的表达差异无统计学意义(P>0.05),β-CAT蛋白的表达差异有统计学意义(P<0.05),两指标在年龄、是否绝经、手术病理分期、组织学分级组的表达差异无统计学意义(P>0.05)。PTEN与β-CAT的蛋白表达呈显著负相关(r=-0.398,P<0.01)。结论 PTEN、β-CAT蛋白的表达与子宫内膜癌的发生相关。PTEN、β-CAT基因突变在不典型增生阶段已经发生,且增殖活性的改变与癌变过程相关,对了解子宫内膜癌的发病机制有重要的参考价值。
Objective To study the expression of PTEN and β-CAT in normal endometrium, endometrial hyperplasia and endometrial carcinoma, and to explore its relationship with the occurrence and development of endometrial carcinoma and its potential as a biological indicator of early canceration. Methods The expressions of PTEN and β-CAT in 26 cases of normal endometrium, 55 cases of endometrial hyperplasia and cases of endometrial carcinoma were studied by immunohistochemical S-P method. Results The positive expression rates of PTEN protein in normal endometrium, endometrial hyperplasia and uterine endometrial adenocarcinoma tissues decreased in descending order, the difference was statistically significant (P <0.01). The positive expression rate of β-CAT protein Followed by increasing, the difference was statistically significant (P <0.01). There was no significant difference in the expression of PTEN protein between different muscle layers (P> 0.05), while the expression of β-CAT protein had statistical significance (P <0.05). The two indexes were different in age, There was no significant difference in the expression of the group (P> 0.05). There was a significant negative correlation between PTEN and β-CAT (r = -0.398, P <0.01). Conclusion The expression of PTEN and β-CAT is related to the occurrence of endometrial carcinoma. PTEN, β-CAT gene mutation in the atypical hyperplasia stage has occurred, and changes in proliferative activity and cancer related to the process of understanding the pathogenesis of endometrial cancer have important reference value.
