目的评价FRET探针定量分型方法对HBV分型的可靠性和在分子流行病学调查中的可行性。方法对随机抽取的38(人)份广西柳州献血者的HBSAg阳性血浆标本,分别进行FRET探针方法分型和S测序分型,比较分型结果。结果使用FRET探针方法的正确分型率为84.21%(32/38),38份标本S区测序得到HLA-B型29例、C型7例及D和G型各1例。结论 FRET探针定量分型灵敏度较高,简便快速,适用于大通量标本分型并能鉴别不同型别毒株混合感染的标本;在分型的同时还可以得到标本病毒载量信息,可以用于HBV的分子流行病调查。
Objective To evaluate the reliability of quantitative typing of FRET probe in HBV genotyping and its feasibility in molecular epidemiological investigation. Methods HBSAg positive plasma samples of 38 (human) donors from Guangxi Liuzhou were randomly selected for FRET probe typing and S sequencing typing, and the typing results were compared. Results The correct typing rate was 84.21% (32/38) using the FRET probe method. In the 38 specimens, 29 cases of HLA-B type, 7 cases of type C and 1 case of type D and G were obtained in 38 specimens. Conclusions Quantitative typing of FRET probe is sensitive, simple and rapid, suitable for typing large samples and capable of identifying mixed infection samples of different genotypes.
