目的:建立知母-黄芪中芒果苷、毛蕊异黄酮苷和毛蕊异黄酮3种有效成分的含量测定方法。方法:采用HPLC,Phenomenex Luna C18(4.8 mm×250 mm,5μm)色谱柱,加预柱(Security Guard,C18,3.0 mm×4.0 mm,Phenomenex);以乙腈-0.2%磷酸溶液为流动相,梯度洗脱,流速1.0 mL.min-1,柱温30℃,检测波长260 nm。结果:芒果苷、毛蕊异黄酮苷和毛蕊异黄酮的线性范围分别为36.733~550.999 ng,33.376~166.88 ng,44.544~222.72 ng,相关系数(R2)均>0.999,平均加样回收率分别为102.47%,102.38%,100.20%,其RSD分别为2.23%,2.86%,2.06%。结论:方法简便快速,重复性良好,结果准确可靠,可用于知母-黄芪中芒果苷、毛蕊异黄酮苷和毛蕊异黄酮的含量测定。
OBJECTIVE: To establish a method for the determination of three effective constituents of mangiferin, calycosin and calycosin in Anemarrhena asphodeloides Astragali. METHODS: HPLC was performed on a Phenomenex Luna C18 (4.8 mm × 250 mm, 5 μm) column with a guard column (C18, 3.0 mm × 4.0 mm, Phenomenex). The mobile phase was acetonitrile-0.2% phosphoric acid Eluted at a flow rate of 1.0 mL.min-1, the column temperature was 30 ℃ and the detection wavelength was 260 nm. Results: The linear ranges of mangiferin, calycosin and calycosin were 36.733-550.999 ng, 33.376-166.88 ng and 44.544-222.72 ng, respectively. The correlation coefficients (R2) were both> 0.999. The average recoveries were 102.47% 102.38% and 100.20% respectively, with RSDs of 2.23%, 2.86% and 2.06% respectively. Conclusion: The method is simple, rapid, reproducible and accurate. It can be used to determine the content of mangiferin, calycosin and calycosin in Radix Astragali.
