目的研究大鼠急性心肌梗死(AMI)后心肌组织中melusin/Akt通路和心功能的动态变化。方法 SPF级雄性SD大鼠38只,随机分为AMI组(结扎冠状动脉左前降支)(n=30)和对照组(开胸不结扎冠状动脉)(n=8),AMI组分为AMI后1、4、6、8周亚组。用Westernblot法检测melusin的表达及Akt磷酸化水平,超声心动图检测心功能的变化,心导管法检查血流动力学的变化,HE染色检测心脏形态学的改变。结果 AMI后早期心肌melusin表达、Akt磷酸化水平均较对照组明显降低,此后逐步升高,第6周达到高峰,并超过对照组,然后下降,第8周的表达水平低于对照组;AMI组心功能(FS%、dp/dtmax)较对照组明显降低,AMI8周组较1、4、6周组显著降低。结论 AMI后心功能的变化和melusin/Akt通路的变化趋势相似,提示melusin/Akt通路可能参与了心梗后大鼠心脏重构和心功能的降低。
Objective To study the dynamic changes of melusin / Akt pathway and cardiac function in myocardial tissue after acute myocardial infarction (AMI) in rats. Methods Thirty-eight SPF male SD rats were randomly divided into AMI group (ligation of the left anterior descending coronary artery) (n = 30) and control group (n = 8) After 1,4,6,8 weeks subgroup. The expression of melusin and phosphorylation of Akt were detected by Western blot. The changes of cardiac function were detected by echocardiography. The changes of hemodynamics were examined by cardiac catheterization. The changes of cardiac morphology were detected by HE staining. Results After AMI, the expression of melusin and Akt phosphorylation were significantly lower than those in the control group, and then gradually increased, reached the peak at the 6th week, and then decreased. The expression level of AMI in the 8th week was lower than that of the control group The cardiac function (FS%, dp / dtmax) was significantly lower than that of the control group. The AMI8 week group was significantly lower than the 1,4,6 week group. Conclusions The change of cardiac function after AMI is similar to that of melusin / Akt pathway, suggesting that melusin / Akt pathway may be involved in cardiac remodeling and cardiac function reduction after AMI.
