目的:建立猪苓药材中麦角甾酮的HPLC含量测定方法。方法:采用甲醇超声提取方法;色谱条件:Inertsil ODS-3(250mm×4.6mm,5μm)柱,流动相为甲醇-水(96∶4),流速1.0mL·min-1,检测波长为350nm,柱温30℃。结果:麦角甾酮在2.5~35μg·mL-1(r=0.9997)范围内与峰面积呈良好的线性关系;麦角甾酮的平均回收率(n=5)为100.5%,RSD为2.5%。结论:建立的方法灵敏、快速、准确,适合作为猪苓药材中麦角甾酮的含量测定,可为猪苓药材质量评价提供可靠的方法。
Objective: To establish a HPLC method for the determination of ergosterone in Polyporus umbellatus. The mobile phase consisted of methanol-water (96: 4), the flow rate was 1.0 mL · min-1 and the detection wavelength was 350 nm. The chromatographic conditions were as follows: Column temperature 30 ℃. RESULTS: The calibration curve of ergosterone showed a good linear relationship with the peak area in the range of 2.5 ~ 35μg · mL-1 (r = 0.9997). The mean recoveries of ergosterone (n = 5) were 100.5% and the RSD was 2.5%. Conclusion: The established method is sensitive, rapid and accurate and suitable for the determination of ergosterone in Polyporus umbellatus, which can provide a reliable method for the quality evaluation of Polyporus umbellatus.
