目的了解婴幼儿下呼吸道细胞实验感染呼吸道合胞病毒后不同时间点受体细胞转录水平,为此类疾病的防治提供指导。方法取婴幼儿下呼吸道细胞进行培养并分为病毒感染组和正常对照组。提取细胞总RNA,进行定性和定量分析,进行RT-PCR检测。结果不同时间点RT-PCR检测正常细胞TLRs(TLR3,TLR4,TLR7和TLR9)的mRNA有一定程度的表达,呼吸道合胞病毒感染细胞TLRs的mRNA随病毒感染时间延长呈上调趋势。结论婴幼儿下呼吸道细胞感染呼吸道合胞病毒后TLR3,TLR4,TLR7和TLR9的基因mRNA转录水平上调,可供婴幼儿下呼吸道感染的防治提供参考。
Objective To understand the transcriptional level of receptor cells at different time points after infection of respiratory syncytial virus in infants and young children’s lower respiratory tract cells and provide guidance for the prevention and treatment of such diseases. Methods Infants and young children lower respiratory tract cells were cultured and divided into viral infection group and normal control group. Total cellular RNA was extracted for qualitative and quantitative analysis, and RT-PCR was performed. Results The mRNA of TLRs (TLR3, TLR4, TLR7 and TLR9) in normal cells was detected by RT-PCR at different time points. The mRNA of TLRs in respiratory syncytial virus infected cells showed an upward trend with the prolongation of virus infection time. Conclusion The mRNA transcription level of TLR3, TLR4, TLR7 and TLR9 in infants with lower respiratory tract infection of respiratory syncytial virus is upregulated, which may provide a reference for the prevention and treatment of lower respiratory tract infection in infants.
