目的探讨β-catenin在结直肠癌(colorectal cancer,CRC)组织的表达及其生物学意义。方法选取2001年2月-2001年9月在北京大学临床肿瘤学院手术治疗的36例CRC患者, 以免疫组织化学S-P法检测β-catenin在癌组织及相应的对照正常组织中的表达,并分析β-catenin的表达与CRC临床病理因素之间的相关性,用逆转录-聚合酶链式反应(RT-PCR)法比较β-catenin mRNA在癌组织及相应正常组织之间的差异。结果83.3%的肿瘤组织β-catenin表达阳性,5.6%的自身对照正常组织表达阳性(P<0.01);β-catenin的表达与肿瘤的肿瘤分化程度及TNM分期之间具有显著相关性(分别为r=-0.880,P=0.009;r=-0.796,P=0.026)。结论β-catenin蛋白表达与TNM分期及肿瘤分化程度之间具有显著相关性。β-catenin蛋白在CRC中的高表达可能并非由于其生成增加,而可能是降解减少所致。
Objective To investigate the expression of β-catenin in colorectal cancer (CRC) and its biological significance. Methods Thirty-six patients with CRC who underwent surgery from Peking University School of Clinical Oncology from February 2001 to September 2001 were recruited. The expression of β-catenin in cancer tissue and corresponding normal tissues was detected by immunohistochemical SP method. β-catenin expression and CRC clinicopathological factors, the difference between β-catenin mRNA and corresponding normal tissues was compared by reverse transcription-polymerase chain reaction (RT-PCR). Results The positive expression of β-catenin was found in 83.3% of the tumors and 5.6% of the normal controls (P <0.01). The expression of β-catenin was positively correlated with the tumor differentiation and TNM stage Significantly correlated (r = -0.880, P = 0.009; r = -0.796, P = 0.026, respectively). Conclusion There is a significant correlation between the expression of β-catenin and TNM stage and tumor differentiation. The high expression of β-catenin protein in CRC may not be due to its increased production, but may be due to the reduction of degradation.
