目的:探讨去甲肾上腺素(noradrenalin,NA)对THP-1巨噬细胞ATP结合盒转运体A1(ATP binding cassette transporter A1,ABCA1)、A类清道夫受体(Scavenger receptorAⅠ,SR-AⅠ)和B型清道夫受体CD36(Cluster of Differentiation 36,CD36)表达的影响。方法:不同浓度NA处理THP-1巨噬细胞24小时,运用逆转录多聚酶链反应(reverse transcription polymerase chain reaction,RT-PCR)检测其ABCA1、SR-AⅠ和CD36的mRNA表达,western-blotting检测ABCA1和SR-AⅠ蛋白的表达。结果:100nmol/L,1μmol/L及10μmol/L的NA作用于THP-1巨噬细胞,引起ABCA1 mRNA和蛋白水平下调(P<0.05),1μmol/L及10μmol/L的NA则升高SR-AⅠ mRNA和蛋白的水平(p<0.05),10pmol/L~10μmol/L的NA作用THP-1巨噬细胞24小时,各组CD36 mRNA表达与对照组比较其差异无统计学意义。结论:应激浓度的NA可能通过影响ABCA1和SR-AⅠ的表达而参与和/或促进AS的发生发展。
Objective: To investigate the effects of noradrenalin (NA) on the expression of ATP binding cassette transporter A1 (ABCA1), Scavenger receptor AⅠ (SR-AⅠ) and THP-1 macrophages B-type scavenger receptor CD36 (Cluster of Differentiation 36, CD36) expression. Methods: THP-1 macrophages were treated with different concentrations of NA for 24 hours. The mRNA expression of ABCA1, SR-AⅠ and CD36 was detected by reverse transcription polymerase chain reaction (RT-PCR) And SR-AI protein expression. Results: The mRNA and protein levels of ABCA1 were down-regulated in THP-1 macrophages with 100nmol / L, 1μmol / L and 10μmol / L NA. (P <0.05). When THP-1 macrophages were treated with 10 pmol / L ~ 10 μmol / L NA for 24 hours, the expression of CD36 mRNA in each group had no significant difference compared with the control group. Conclusions: NA at stress concentration may participate in and / or promote the development of AS by affecting the expression of ABCA1 and SR-AI.
