目的探讨阻燃剂得克隆的环境激素作用。方法将去势Wistar雌鼠随机分为6组,去卵巢(模型)组、假手术组、得克隆低、中、高剂量组(25、50、100 mg/kg),阳性对照组(雌二醇50 mg/kg);连续灌胃14 d,第15 d处死,收集血清、摘取子宫称重并冷冻保存,通过电化学发光免疫法检测大鼠血清甲状腺激素(FT3、FT4、TSH)含量,采用Real-time RT PCR法检测子宫雌激素受体α(Erα)mRNA表达水平。结果与假手术组比较,模型组大鼠子宫重量[(0.112±0.035)g]与子宫/体重比值(0.072±0.030)明显降低,子宫内ErαmRNA表达水平明显下降(P<0.05);与模型组比较,高剂量得克隆组大鼠子宫重量[(0.419±0.059)g]与子宫/体重比值(0.186±0.143)明显上升,子宫内ErαmRNA表达量明显增加,差异有统计学意义(P<0.05);各组血清甲状腺激素水平无明显变化。结论得克隆在体内具有雌激素样作用,但对血清甲状腺激素无影响。
Objective To investigate the environmental hormone effect of flame retardant cloning. Methods The castrated Wistar female rats were randomly divided into 6 groups: ovariectomized group, sham operation group, low, middle and high dose of cloned group (25, 50, 100 mg / kg), positive control group Alcohol 50 mg / kg). The rats were sacrificed on the 15th day after continuous gavage. Serum was harvested and the uterus was harvested for weighing and cryopreservation. Conclusion Clone has estrogen-like effect in the body, but no effect on serum thyroid hormone.
