目的探讨枸杞多糖(LBP)对小鼠宫颈癌细胞增殖的影响及其机制。方法 MTT法检测不同浓度LBP作用48h后对体外培养的宫颈癌U14细胞的抑制率。ELISA法检测LBP 1000μg/ml作用48h后U14细胞分泌IL-12及IL-10的水平。构建20只C57BL/6J小鼠U14细胞皮下成瘤模型,随机均分为两组,实验组每日灌胃LBP 10mg/kg,对照组予等量生理盐水。给药4周后ELISA法检测两组小鼠血清IL-12及IL-10的水平;计算小鼠瘤质量、体积、胸腺指数和脾脏指数;免疫组化法检测瘤块中Ki-67的表达。结果 LBP呈剂量依赖性地抑制U14细胞的生长。LBP 1000μg/ml作用后,U14细胞IL-12分泌增加,IL-10分泌减少(P<0.01)。与对照组比较,实验组小鼠血清IL-12水平、胸腺和脾脏指数升高,血清IL-10水平、瘤质量、体积及Ki-67表达量降低(P<0.01)。结论 LBP能够通过上调IL-12及下调IL-10的水平达到抗小鼠宫颈癌细胞增殖的作用。
Objective To investigate the effect of LBP on the proliferation of mouse cervical cancer cells and its mechanism. Methods MTT assay was used to detect the inhibitory rate of U14 cells cultured in vitro after different concentrations of LBP for 48 hours. The levels of IL-12 and IL-10 secreted by U14 cells after LBP 1000μg / ml 48h were detected by ELISA. Twenty adult C57BL / 6J mice were inoculated subcutaneously into U14 cells and randomly divided into two groups. The experimental group received intragastric administration of LBP 10 mg / kg daily and the control group received the same volume of normal saline. After 4 weeks of administration, the levels of IL-12 and IL-10 in serum of the two groups were detected by ELISA; the tumor mass, volume, thymus index and spleen index were calculated; the expression of Ki-67 in tumor mass was detected by immunohistochemistry . Results LBP dose-dependently inhibited the growth of U14 cells. After LBP 1000μg / ml, U14 cells increased IL-12 secretion and IL-10 secretion decreased (P <0.01). Compared with the control group, serum IL-12 level, thymus and spleen index, serum IL-10 level, tumor mass, volume and Ki-67 expression in the experimental group decreased (P <0.01). Conclusion LBP can up-regulate the level of IL-12 and down-regulate the level of IL-10 to inhibit the proliferation of mouse cervical cancer cells.
