背景:信号传导及转录活化因子3(signaltransducerandactivatoroftran-scription3,STAT3)蛋白广泛表达于不同类型的细胞和组织中,是重要的信号转导及转录级联成分,参与调控细胞的增殖分化等多种生理功能。但是在胚胎肝脏发育中是否有STAT3的表达,STAT3是否参与了胚胎肝脏中期发育过程,目前国内外尚无文献报道。目的:通过研究STAT3蛋白在小鼠胚胎肝脏发育中的表达,试图揭示STAT3信号转导通路在胚胎肝脏发育过程中的作用和可能机制。设计:以STAT3蛋白为研究对象随机对照的基础研究。单位:一所军医大学的组胚教研室。材料:本实验于2004-01/2004-06在解放军第三军医大学组胚教研室的实验室完成。KM种小鼠(清洁级)由解放军第三军医大学动物中心提供。随机取雄鼠10只,雌鼠20只,体质量25~30g。方法:分别取交配后13.5d(n=8)、交配后14.5d(n=9)、交配后15.5d(n=7)鼠胚做冰冻连续切片。每2张切片中取1张用0.01mol/LPBS代替STAT3一抗设阴性对照实验。利用免疫细胞化学技术和免疫荧光细胞化学技术显示STAT3蛋白在小鼠胚胎肝脏发育中的表达。主要观察指标:鼠胚冰冻连续切片STAT3蛋白的表达情况。结果:在孕13.5d(E13.5d)鼠胚中,STAT3蛋白在肝细胞中的表达较强,孕14.5d(E14.5d)和孕15.5d(E15.5d)STAT3蛋白在肝脏中的表?
BACKGROUND: Signal transduction and activator of transcription 3 (STAT3) proteins are widely expressed in different types of cells and tissues and are important signal transduction and transcriptional cascades. They are involved in the regulation of cell proliferation and differentiation and many other physiological Features. However, whether STAT3 is expressed in embryonic liver development or not, STAT3 is involved in the mid-term development of embryonic liver. Objective: To investigate the role of STAT3 signal transduction pathway in the development of embryonic liver and its possible mechanism by studying the expression of STAT3 protein in mouse embryonic liver. Design: The basic research of randomized controlled study of STAT3 protein. Unit: a group of embryos in a military medical university. MATERIALS: The experiment was performed in the laboratory of embryos of the Third Military Medical University of PLA from January 2004 to June 2004. KM species of mice (clean level) by the Third Military Medical University Animal Center. Ten male rats were randomly selected, 20 female rats, body weight 25 ~ 30g. Methods: The frozen embryos were harvested at 13.5d (n = 8), 14.5d after mating (n = 9) and 15.5d after mating (n = 7) respectively. Take a negative control experiment with 0.01 mol / L LPBS instead of STAT3 primary antibody in every two sections. Immunocytochemistry and immunofluorescence cytochemistry were used to show the expression of STAT3 protein in mouse embryonic liver. MAIN OUTCOME MEASURES: STAT3 protein expression in frozen sections of rat embryos. Results: STAT3 protein was strongly expressed in hepatocytes in embryonic 13.5d (E13.5d) embryos, and the expression of STAT3 protein in liver at 14.5d (E14.5d) and 15.5d (E15.5d) ?
