目的探究采用超声介导靶向微泡[自制,含造影剂(全氟显)、血管内皮生长因子_(165)c脱氧核糖核酸(VEGF_(165)c DNA)、抗细胞间粘附因子(ICAM-1)单克隆抗体]治疗脑梗死白兔的动物实验效果,同时观察VEGF_(165)c DNA基因转染对改善受损血管内膜的作用。方法选取60只新西兰大白兔,通过高脂饮食制作脑梗死模型,随机分为3组,分别采用超声介导靶向微泡(A组)、爱通立(B组)、生理盐水(对照组)进行治疗,观察各组白兔治疗后血管再通情况、梗死部位坏死区中性粒细胞浸润情况、梗死区质量比重。结果治疗后A组及B组血管再通情况优于对照组(P<0.05),但A组与B组血管再通情况比较差异无统计学意义(P>0.05);治疗后A组梗死部位坏死区中性粒细胞平均计数明显少于B组及对照组(P<0.05),但B组与对照组梗死部位坏死区中性粒细胞平均计数对比差异无统计学意义(P>0.05);治疗后A组梗死区质量占比低于B组及对照组(P<0.05),且B组梗死区质量占比低于对照组(P<0.05)。结论超声联合靶向微泡介导脑梗死治疗的动物实验VEGF_(165)c DNA基因转染治疗效果较好,能改善血管再通情况,减轻梗死区域的炎性反应,减少梗死区质量占比,有利于治疗脑梗死。
OBJECTIVE: To investigate the effect of ultrasound-mediated microbubble targeting (VEGF-165) c DNA, anti-cell adhesion molecule ICAM-1) monoclonal antibody] in the treatment of cerebral infarction in rabbits, and to observe the effect of VEGF165c DNA gene transfection on improving the damaged vascular intima. Methods Sixty New Zealand white rabbits were selected to make cerebral infarction model by high-fat diet. The models were randomly divided into three groups: ultrasound-guided targeted microbubbles (A group), Aitongli (B group), normal saline ) For treatment. The recanalization of the rabbits in each group was observed, the neutrophil infiltration in the necrotic area of infarction and the weight of infarct area were observed. Results The vascular recanalization in group A and group B was better than that in control group (P <0.05), but there was no significant difference between group A and group B (P> 0.05). After treatment, the infarct size in group A Neutrophil count in necrotic area was significantly lower than that in B group and control group (P <0.05). However, there was no significant difference in the average number of neutrophils in necrotic area between B group and control group (P> 0.05). The quality of infarction mass in group A was lower than that in group B and control group (P <0.05), and the proportion of mass in group B was lower than that in control group (P <0.05). CONCLUSIONS: Ultrasound combined with VEGF-C (165) c DNA transfection in mice treated with microbubble-targeted cerebral infarction has a good effect in improving the vascular recanalization, reducing the inflammatory response in the infarct area and reducing the mass ratio of infarct area , Is conducive to the treatment of cerebral infarction.
