目的:探讨瘦素对人脐静脉内皮细胞(ECV-304)表达CD40和NF-κB的影响。方法:人脐静脉内皮细胞培养及分组(6h作用组和24h作用组),用10μg/L、30μg/L、50μg/L的瘦素及50μg/L瘦素加上NF-κB阻断剂PDTC处理;免疫组织化学方法检测CD40和NF-κB的表达。结果:CD40为棕黄色颗粒,主要分布于细胞膜和细胞质;在10μg/L的瘦素处理后,随着时间的延长,CD40表达水平不变,在30μg/L、50μg/L的瘦素处理后,随着时间的延长,CD40表达水平升高,在相同的作用时间下,随着浓度的增高,CD40表达水平升高,差异有统计学意义。NF-κB(P65)为棕黄色颗粒,主要表达于细胞质,在10μg/L的瘦素处理后,随着时间的延长,NF-κB(P65)表达水平不变,在30μg/L、50μg/L的瘦素处理后,随着时间的延长,NF-κB(P65)表达水平升高;在相同的作用时间下,随着浓度的增高,NF-κB(P65)表达水平升高,差异有统计学意义;按20μl/105细胞加入CD40单克隆抗体处理6h后,NF-κB(P65)表达水平下降,经统计学检验,CD40和NF-κB(P65)的表达有相关性。结论:高浓度瘦素可以诱导脐静脉内皮细胞同时表达CD40和NF-κB(P65),同时,CD40表达被阻止后,NF-κB(P65)表达水平降低,说明二者的表达有相关性,CD40和NF-κB(P65)可能共同参与了高瘦素血症引发的动脉粥样硬化的发生和发展。
Objective: To investigate the effect of leptin on the expression of CD40 and NF-κB in human umbilical vein endothelial cells (ECV-304). CONCLUSION: High concentration of leptin can induce the expression of CD40 and NF-κB (P65) in human umbilical vein endothelial cells. Meanwhile, the expression of NF-κB (P65) is decreased after the expression of CD40 is blocked, CD40 and NF-κB (P65) may participate in the occurrence and development of atherosclerosis induced by hyperlipidemia.
