目的研究EphB4受体和缺氧诱导因子-1α(HIF-1α)蛋白在胃癌组织中的表达,并探讨其生物学意义及相互关系。方法应用免疫组化法检测74例胃癌组织(胃癌组)和13例正常胃黏膜组织(对照组)中EphB4、HIF-1α的表达,并分析其表达与胃癌临床病理因素之间的关系。结果胃癌组中EphB4和HIF-1α的高表达率分别为62.16%(46/74)和52.70%(39/74),在对照组中,EphB4和HIF-1α的高表达率分别为15.38%(2/13)和0%(0/13)。胃癌组织中EphB4和HIF-1α的表达显著高于正常胃黏膜组织(P<0.01)。EphB4、HIF-1α的表达与胃癌的组织学分级及临床病理分期关系密切(P<0.05),而与年龄、性别、肿瘤直径、淋巴结转移、远处转移无关(P>0.05)。与HIF-1α不同,EphB4的表达还与肿瘤浸润深度有关(P<0.05)。此外,EphB4与HIF-1α的表达成显著正相关(r=0.2091,P<0.01)。结论在胃癌中EphB4、HIF-1α均高表达,其表达程度与胃癌的恶性程度有关,而且EphB4表达与HIF-1α的表达呈正相关。过表达的EphB4、HIF-1α在胃癌恶性进展中发挥重要作用。
Objective To study the expression of EphB4 receptor and hypoxia inducible factor-1α (HIF-1α) protein in gastric carcinoma and to explore its biological significance and relationship. Methods The expressions of EphB4 and HIF-1α in 74 cases of gastric cancer (gastric cancer group) and 13 cases of normal gastric mucosa (control group) were detected by immunohistochemistry. The relationship between the expression of EphB4 and HIF-1α and clinicopathological factors was analyzed. Results The high expression rates of EphB4 and HIF-1α in gastric cancer group were 62.16% (46/74) and 52.70% (39/74) respectively. In the control group, the high expression rates of EphB4 and HIF-1α were 15.38% 2/13) and 0% (0/13). The expression of EphB4 and HIF-1α in gastric cancer tissues was significantly higher than that in normal gastric mucosa tissues (P <0.01). The expressions of EphB4 and HIF-1αwere closely related to histological grade and clinicopathologic stage (P <0.05), but not to age, gender, tumor diameter, lymph node metastasis and distant metastasis (P> 0.05). Unlike HIF-1α, the expression of EphB4 was also related to the depth of tumor invasion (P <0.05). In addition, there was a significant positive correlation between EphB4 and HIF-1α expression (r = 0.2091, P <0.01). Conclusions EphB4 and HIF-1α are both highly expressed in gastric cancer. The expression of EphB4 and HIF-1α is correlated with the degree of malignancy of gastric cancer, and the expression of EphB4 is positively correlated with the expression of HIF-1α. Overexpression of EphB4 and HIF-1α play an important role in the malignant progression of gastric cancer.
