目的检测Notch家族成员在小细胞肺癌(SCLC)组织中基因及蛋白表达水平,揭示Notch家族成员表达与小细胞肺癌发生、发展的关系。方法收集小细胞肺癌新鲜组织,迅速放入液氮中,RT-PCR技术检测Notch-1、Notch-2、Notch-3、Notch-4(N1、N2、N3、N4)的基因表达水平;组织切片行免疫组织化学检测N1、N2、N3、N4蛋白表达水平。所有患者术后均行常规化疗药物治疗。结果 10例患者中N1、N2、N3、N4表达程度并不一致,有2例N2表达降低,还有2例N3表达降低。患者预后也不一样,N2表达降低的患者其预后好于表达高的患者,5年生存率达100%。结论 Notch的表达与小细胞肺癌预后可能有关,低表达者预后更好。
Objective To detect the expression of Notch family members in small cell lung cancer (SCLC) tissues and to explore the relationship between the expression of Notch family members and the occurrence and development of small cell lung cancer. Methods Fresh tissue of small cell lung cancer was collected and rapidly filled into liquid nitrogen. The gene expression levels of Notch-1, Notch-2, Notch-3 and Notch-4 (N1, N2, N3 and N4) were detected by RT- The expression of N1, N2, N3 and N4 protein was detected by immunohistochemistry. All patients underwent conventional chemotherapy after surgery. Results The expression of N1, N2, N3 and N4 in 10 patients was not consistent. The expression of N2 was decreased in 2 cases and decreased in 2 cases. Patients with different prognosis, N2 expression decreased in patients with better prognosis than those with high expression, 5-year survival rate of 100%. Conclusion The expression of Notch may be related to the prognosis of small cell lung cancer, and the prognosis of low expression is better.
