目的:探讨β-神经生长因子(β-NGF)对大鼠内皮祖细胞(EPCs)内皮型一氧化氮合酶(eNOS)及一氧化氮(NO)合成的影响。方法:将β-NGF重组腺病毒(Ad-β-NGF)及其阴性对照感染至大鼠内皮祖细胞,同时设置空白对照,分别于24h、48h及72h进行检测。用qPCR及Western blot分别在mRNA和蛋白水平检测不同组eNOS表达情况,硝酸还原酶法测定培养液中NO的水平。结果:Ad-NGF感染组eNOS表达及NO合成明显高于对照组,差异有统计学意义(P<0.05),并随感染时间的延长而增加。结论:β-NGF能促进内皮祖细胞eNOS表达及NO合成,有利于内皮祖细胞发挥功能。
Objective: To investigate the effect of β-NGF on the synthesis of endothelial nitric oxide synthase (eNOS) and nitric oxide (NO) in rat endothelial progenitor cells (EPCs). Methods: Ad-β-NGF recombinant adenovirus (Ad-β-NGF) and its negative control were transfected into endothelial progenitor cells of rats. Blank control was set at 24 h, 48 h and 72 h respectively. The expression of eNOS in different groups was detected by qPCR and Western blot at mRNA and protein levels, respectively. Nitric acid reductase was used to determine the level of NO in culture media. Results: The expression of eNOS and NO synthesis in Ad-NGF group were significantly higher than those in control group (P <0.05), and increased with the prolongation of infection time. CONCLUSION: β-NGF can promote the eNOS expression and NO synthesis of endothelial progenitor cells, which is beneficial to the function of endothelial progenitor cells.
