为探讨胃内局部环境中胃酸、胃蛋白酶等因素对胃安素(TP)药效的影响,对经人工胃液处理前后三批不同时间制备的TP进行了活力检测及动物模型试验。结果:1.未经人工胃液处理,三批TP(100mg/L)均显示较强的促3T3成纤维细胞增殖活力,而以920620及930713批TP偏低。2.经人工胃液处理0.5、1、2小时,930713、940528二批TP均有活力,以940528批活力较高,即经处理1小时、稀释度为2mg/L时,活力比值X>2.0;而920620批未能测出活力(X<2.0)。3.动物模型实验结果:与对照组比较,930713、940528批TP组溃疡面积在喂药10天后均缩小,具高度显著性差异(P<0.01);920620批TP疗效则不显著(P>0.05)。结果提示,人工胃液条件下pH、胃蛋白酶对TP活力有一定影响,并与其消化作用及TP储存时间长短相关。活力检测与动物模型试验结果相符,并具一定的平行性。
In order to investigate the effect of gastric acid and pepsin on the pharmacodynamics of TP, the vitality test and animal model test of TP prepared by three groups of different time before and after artificial gastric juice treatment were conducted. Three groups of TP (100mg / L) showed a strong activity of promoting the proliferation of 3T3 fibroblasts without artificial gastric juice treatment, while TP was lower with 920620 and 930713 batches. 2, 0.5,1,2 hours after artificial gastric juice treatment, 930713,940,528 two batches of TP are viable to 940528 batch of high activity, that is treated for 1 hour, the dilution of 2mg / L, the activity ratio X> 2.0; The 920,620 batch failed to measure vitality (X <2.0). Animal model experimental results: Compared with the control group, the ulcer area of 930713 and 940528 batches of TP group were all reduced after 10 days of feeding, with a highly significant difference (P <0.01); while the 920620 batches of TP had no significant effect (P> 0.05 ). The results suggest that pH and pepsin have an impact on the activity of TP under the condition of artificial gastric juice, and are related to the digestion and the storage time of TP. Vitality test and animal model test results are consistent with a certain degree of parallelism.
