目的通过检测颌下腺细胞凋亡及Fas/FasL表达程度,探讨路氏滋燥汤治疗干燥综合征的作用机制。方法采用白百破疫苗加完全弗氏佐剂及同种鼠颌下腺抗原联合诱导干燥综合征动物模型。治疗组小鼠以路氏滋燥汤灌胃,空白组和模型组给予生理盐水灌胃。两周后处死动物,流式细胞仪检测各组小鼠颌下腺组织细胞凋亡率、免疫组化法测定各组小鼠颌下腺中Fas/FasL水平。结果相对于模型组,2个治疗组小鼠颌下腺细胞凋亡率及Fas/FasL表达程度明显降低。结论路氏滋燥汤可降低诱导干燥小鼠颌下腺细胞凋亡率及Fas/FasL表达程度,是其治疗干燥综合征的作用机理之一。
Objective To detect the apoptosis of submandibular gland cells and the expression of Fas / FasL and to explore the mechanism of Luoshi Zizania decoction in treating Sjogren’s syndrome. Methods The animal model of Sjogren ’s syndrome was induced by the combination of Pertussis Vaccine plus complete Freund’s adjuvant and the same rat submandibular gland antigen. The mice in the treatment group were given Rouzizhixiao decoction intragastrically, while the blank group and model group were given normal saline. Animals were sacrificed two weeks later, apoptosis rate of submandibular gland cells in each group was detected by flow cytometry, and Fas / FasL level in submandibular gland of mice in each group was determined by immunohistochemical method. Results Compared with the model group, the apoptotic rates and the expression of Fas / FasL in the submandibular gland cells of the two treatment groups were significantly decreased. Conclusion Luxbu Zizania Decoction can reduce the rate of apoptosis and the expression of Fas / FasL in the submandibular gland cells induced by dryness in mice, which is one of its mechanism of action in the treatment of Sjogren’s syndrome.
