目的初步探讨宫颈癌中转移抑制基因1(MTSS1)与Shh信号通路间的关系。方法取2011年12月至2015年10月期间采集的90例宫颈组织,应用实时荧光定量PCR(q-PCR)检测正常宫颈组(A组)、早期宫颈癌组(B组)及中晚期宫颈癌组(C组)中MTSS1基因及Shh信号通路相关基因的表达水平;Western印迹法及免疫组化检测MTSS1及Gli1蛋白表达情况。结果 q-PCR结果显示,随着临床分期的升高,MTSS1、Ptch、Smo及Gli1基因表达水平逐渐升高,其中C组中4个基因mRNA表达量均最高(P<0.01);Western印迹结果显示,C组MTSS1蛋白表达率明显高于B组及A组(P<0.05);免疫组化检测MTSS1及Gli1蛋白在B和C组较A组中的表达增强,其中C组中两者多为强阳性表达。结论宫颈癌发生发展中MTSS1的表达与Shh信号调控相一致,均随癌症发展表达增强,可能同为预测宫颈癌发生发展的指标与潜在的治疗靶点。
Objective To investigate the relationship between metastasis suppressor gene 1 (MTSS1) and Shh signaling pathway in cervical cancer. Methods Totally 90 cases of cervical tissue collected from December 2011 to October 2015 were enrolled in this study. Normal cervical group (group A), early cervical cancer group (group B) and middle and late cervical cancer were detected by real-time quantitative PCR (q- The expression of MTSS1 and Shh signal pathway related genes in cancer group (C group). The expression of MTSS1 and Gli1 protein were detected by Western blotting and immunohistochemistry. Results The q-PCR results showed that the expression levels of MTSS1, Ptch, Smo and Gli1 increased with the increase of clinical stage, of which the mRNA expression of four genes in group C was the highest (P <0.01). Western blot results The results showed that the expression of MTSS1 protein in group C was significantly higher than that in group B and group A (P <0.05). The expression of MTSS1 and Gli1 protein in group B and C were higher than group A Strong positive expression. Conclusion The expression of MTSS1 in cervical carcinogenesis is consistent with the regulation of Shh signaling. The expression of MTSS1 is closely related to the development and progression of cervical cancer. It may be an index and a potential therapeutic target for predicting the occurrence and development of cervical cancer.
