目的:探寻祛白脂组方中影响小鼠B16黑素瘤细胞代谢的因素。方法:以小鼠B16细胞增殖活性、黑素生成量、酪氨酸酶活性为指标,以8-甲氧补骨脂素(8-MOP)浓度和辅药为考察因素,选用3×2析因设计研究不同组方祛白脂对小鼠B16黑素瘤细胞代谢的影响,用SPSSl 3.0软件辅助数据分析。结果:8-MOP 10μmol·L-1处理组小鼠B16黑素瘤细胞增殖能力明显高于其他组(P<0.01),8-MOP 50μmol·L-1处理组小鼠B16黑素瘤细胞黑素含量、酪氨酸酶活性明显高于其他组(P<0.01);加入辅药小鼠B16黑素瘤细胞增殖能力、黑素含量、酪氨酸酶活性显著高于不加辅药组(P<0.01);8-MOP浓度和辅药处理的交互作用对小鼠B16黑素瘤细胞增殖能力、黑素含量、酪氨酸酶活性有显著影响(P<0.01)。结论:祛白脂中8-MOP浓度和辅药是影响小鼠B16黑素瘤细胞代谢的主要因素。
Objective: To explore the factors that affect the metabolism of B16 melanoma cells in mice. Methods: The proliferation activity of B16 cells, melanogenesis and tyrosinase activity of mouse B16 cells were used as indexes to investigate the effect of 8-MOP concentration and adjuvant on the B16 cell proliferation. Due to the design and study of different groups of anti-Bai lipid on mouse B16 melanoma cell metabolism, with SPSSl 3.0 software aided data analysis. Results: The proliferation of B16 melanoma cells in 8-MOP 10μmol·L-1 group was significantly higher than that in other groups (P <0.01). The B16 melanoma cells in 8-MOP 50μmol·L-1 treated group (P <0.01). The proliferation, melanin content and tyrosinase activity of B16 melanoma cells in adjuvant-treated mice were significantly higher than those in the untreated group (P <0.01) P <0.01). The interaction of 8-MOP and adjuvant treatment had significant effects on the proliferation, melanin content and tyrosinase activity of B16 melanoma cells (P <0.01). Conclusion: The concentration of 8-MOP and the adjuvant in Qu Bai shengzhi are the main factors affecting the metabolism of B16 melanoma cells in mice.
