目的观察表皮生长因子受体(EGFR)单克隆抗体对人肺癌细胞体外增殖的作用,探讨其在肺癌抗体生物治疗中的应用价值。方法应用杂交瘤技术制备EGFR单克隆抗体并以噻唑蓝(MTT)比色法检测EGFR单克隆抗体对人肺癌细胞体外增殖能力的影响。结果EGFR单克隆抗体对人肺腺癌SPCA1和A549细胞体外增殖有显著的抑制作用,但对小细胞肺癌SH77和NCIH446细胞的生长无明显影响。抗体浓度为15μg/ml时,SPCA1和A549细胞的生长率仅为对照组的6%和12%。结论EGFR单克隆抗体在肺腺癌的生物治疗中具有潜在的应用价值
Objective To observe the effect of epidermal growth factor receptor (EGFR) monoclonal antibody on the proliferation of human lung cancer cells in vitro and explore its application value in the biological treatment of lung cancer antibodies. Methods The monoclonal antibody against EGFR was prepared by hybridoma technique and the effect of EGFR monoclonal antibody on the proliferation of human lung cancer cells in vitro was detected by MTT assay. RESULTS: EGFR monoclonal antibody significantly inhibited the proliferation of human lung adenocarcinoma SPCA 1 and A549 cells in vitro, but had no significant effect on the growth of small cell lung cancer SH 77 and NCI H446 cells. When the antibody concentration was 15 μg/ml, the growth rate of SPCA 1 and A549 cells was only 6% and 12% of the control group. Conclusion EGFR monoclonal antibody has potential application value in the biological treatment of lung adenocarcinoma
