目的研究生姜中主要活性成分姜酚的分离方法。方法选用D4020作为固定相,乙醇水溶液作为流动相,从溶剂萃取法得到的姜油树脂中分离姜酚。采用紫外分光光度法(ultraviolet,UV)与高效液相色谱法(high performance liquid chromatography,HPLC)检测分离纯化过程,讨论柱高、流速、上样量对姜酚分离效果的影响,确定最佳工艺条件。结果柱层析法分离姜酚的实验条件:柱高为25 cm,洗脱速度1 mL/min,上样量100 mL。两次纯化后姜酚纯度可以达到85.24%。结论该方法可以提高姜酚的纯度。
Objective To study the method for the separation of gingerol, the main active ingredient in ginger. Methods D4020 was used as the stationary phase and ethanol was used as the mobile phase to separate gingerol from ginger oleoresin obtained by solvent extraction. The separation and purification process was examined by ultraviolet (UV) and high performance liquid chromatography (HPLC). The effects of column height, flow rate and sample volume on the separation of gingerol were discussed, and the optimum process condition. Results The experimental conditions for separation of gingerol by column chromatography were as follows: column height 25 cm, elution speed 1 mL / min, sample volume 100 mL. Twice purified gingerol purity can reach 85.24%. Conclusion This method can improve the purity of gingerol.
