In 1975 Köhleh and Milstein obtained hybridomas by fusion of mouse myeloma cells and spleen cells immunized with sheep erythrocytes. Such hybridoma cells can be continuously cultured in vitro and produce large amounts of homologous antibodies. The hybridomas obtained are screened to obtain clonal cell lines that produce antibodies against a single antigenic determinant. The resulting monoclonal antibodies provide a very specific and sensitive rod for the detection of antigens. Antibodies that can not be detected by commonly prepared antibodies can be detected.
