目的:探讨慢性乙肝患者CD25、CD25 mRNA表达水平及其临床意义。方法:选择249例典型慢性乙肝患者,用生物素-链霉亲和素(BSA)法和Real-tim e PCR法分别检测PHA诱导前后患者PBMC CD25和CD25 mRNA表达水平;用PCR法检测患者HBV-DNA载量。结果:慢性乙肝患者静息态和诱导态CD25、CD25 mRNA表达水平与正常对照组相比均降低,差异有显著性(P<0.05或P<0.01)。慢性乙肝、肝硬化患者CD25和CD25 mRNA的表达水平不完全相同,但差异无显著性(P>0.05)。结论:乙肝患者存在一定程度的免疫功能低下,CD25、CD25 mRNA表达水平降低,其表达水平与病情活跃程度、病毒复制水平相关。
Objective: To investigate the expression of CD25 and CD25 mRNA in patients with chronic hepatitis B and its clinical significance. Methods: A total of 249 patients with chronic hepatitis B were enrolled in this study. The expression of CD25 and CD25 mRNA in peripheral blood mononuclear cells (PBMCs) before and after PHA induction were detected by Real-time PCR and biotin-streptavidin (BSA) DNA load. Results: The levels of resting and induced CD25 and CD25 mRNA in chronic hepatitis B patients were significantly lower than those in normal controls (P <0.05 or P <0.01). The expression levels of CD25 and CD25 mRNA in patients with chronic hepatitis B and cirrhosis were not identical, but the difference was not significant (P> 0.05). Conclusion: There is a certain degree of immunocompromise in hepatitis B patients, and the expression of CD25 and CD25 mRNA is decreased. The expression of CD25 and CD25 mRNA is correlated with the degree of active disease and the level of virus replication.
