目的观察人成骨肉瘤MG-63细胞和正常人成骨细胞(humanosteoblast-likecellsHOB)胰岛素受体底物(insulinreceptorsubstrateIRS)的表达。方法半定量RT-PCR检测IRSmRNA表达,免疫共沉淀检测IRS蛋白表达。结果人成骨肉瘤MG-63细胞和正常人成骨细胞均有IRS-1、-2、-3、-4mRNA和蛋白质的表达,以IRS-1的表达丰度最高,IRS-4表达丰度最低。其中IRSmRNA和蛋白质在MG-63细胞中的表达丰度较在正常人成骨细胞中的表达丰度稍高。结论人成骨肉瘤MG-63细胞和正常人成骨细胞均可表达IRSmRNA和蛋白质。且IRS不同成员在同一细胞中的表达丰度不同。
Objective To observe the expression of human osteoblastic sarcoma MG-63 cells and normal human osteoblast-like cells (insulin receptor substrate insulin receptor substrate). Methods IRS mRNA expression was detected by semi-quantitative RT-PCR and IRS protein expression was detected by co-immunoprecipitation. Results The expression of IRS-1, -2, -3, -4 mRNA and protein in both human osteosarcoma MG-63 cells and normal human osteoblasts was the highest, with the highest expression of IRS-1 and the expression of IRS-4 lowest. Among them, the expression abundance of IRS mRNA and protein in MG-63 cells was slightly higher than that in normal human osteoblasts. Conclusion Human osteosarcoma MG-63 cells and normal human osteoblasts can express IRS mRNA and protein. And different members of IRS in the same cell expression abundance.
